Ha tag antibody assays using ChIP-on-chip technology

The Ha tag antibody is one of a number of antibodies used for ChIP (Chromatin Immunoprecipitation) and ChIP-on-chip assays. ChIP-on-chip is a high throughput assay, combining ChIP with microarray technology. It identifies and analyses DNA-binding sites of histones, transcription factors and other proteins, on a genome-wide basis.
While ChIP dates from the 1980s, ChIP-on-chip is far more recent, being developed around 2001. The process involves five stages: Crosslinking of DNA with proteins of interest; fragmenting of DNA by sonification; ChIP of DNA-bound proteins with relevant antibodies; DNA purification, and hybridization with DNA microarrays.
A number of companies now produce ChIP-grade antibodies. These are antibodies which have been proven to successfully immunoprecipitate cross-linked chromatin. However, although there are a large number of ChIP-grade reagents available, only a small selection are suitable for ChIP-on-chip experiments. This is because the antibodies must be highly specific to their target epitopes under both dry-lab (fixed) and wet-lab (free in solution) conditions.
The Ha tag antibody is one of several for which ChIP-grade versions have been developed, which has proven useful for ChIP-on-chip assays. Where a protein of interest is proving problematical, it can often be fused to an Ha tag. The tagged protein is then recognised by the ChIP-grade Ha tag antibody. Proteins tagged in this way are usually associated with chromatin function. They include histones and modified histones; transcription factors and members of the ORC (origin recognition complex.)
